EXPRESSION OF THE RECOMBINANT HUMAN EXONUCLEASE1 (hExo1) USING BACULOVIRUS EXPRESSION SYSTEM

Abstract
Human exonuclease1 (hExo1) acts directly in diverse DNA processing events, including replication, mismatch repair (MMR) and double strand break repair (DSBR). It was also recently described to function as damage sensor and apoptosis inducer when DNA damage
occur. The aim of this study is to express the recombinant hExo1 protein from insect cells. Recombinant hExo1 protein was functionally expressed in insect High Five™ cells by the BAC-TO-BAC®baculovirus expression system. Recombinant hExo1 was purified to near homogeneity by affinity chromatography using heparin column. A yield of about 200 μg of the hExo1 protein was purified from 500 mL of infected High Five™ cells. The purified protein was analysed by SDS/PAGE, which revealed that the hExo1 protein was isolated in its monomeric form.
